summary: CRISPR/Cas9 was used to engineer the D465N mutation (GACAAC) into the mouse Snx1 gene (note that human and mouse numbering differ by a single amino acid). This models a common missense variant (rs1802376) in SNX1 which has been associated with late-onset AD in multiple independent cohorts. This is expressed on a sensitized model also expressing a humanized APOE4 knock-in and a CRISPR/cas9-generated R47H point mutation in the mouse Trem2 gene.
